Unraveling the Benefits of Bacillus subtilis DSM 29784 Poultry Probiotic through Its Secreted Metabolites: An In Vitro Approach

The study investigates the probiotic Bacillus subtilis 29784 (Bs29784) and its impact on chicken intestinal health via its secreted metabolites: hypoxanthine (HPX), niacin (NIA), and pantothenate (PTH). Using in vitro models, the research reveals how these metabolites influence three key aspects of intestinal resilience: immune response, intestinal barrier, and microbiota. Key findings include:

Vegetative Cells vs. Spores : Bs29784 vegetative cells are more effective in reducing inflammation compared to spores, suggesting the benefits are linked to metabolic activity.

Metabolite Effects:

  • PTH and NIA: Reduce pro-inflammatory pathway activation (AP-1 and NF-κB).

  • HPX enhances mucin production by upregulating MUC2 expression.

  • HPX, NIA, and PTH: Increase cell proliferation.

  • PTH and HPX: Improve epithelial resilience by limiting increased permeability during inflammation.

Fermentation Profiles:

  • In cecal fermentations, NIA increases acetate, HPX increases butyrate, and PTH increases acetate, butyrate, and propionate.

  • In ileal fermentations, PTH increases butyrate.

  • All metabolites modulate microbiota, resulting in varied fermentation patterns.

Overall, Bs29784 positively influences intestinal health by acting on the three lines of resilience: immune response, intestinal barrier integrity, and microbiota composition through its secreted metabolites.
